- Why 4/10
- The defining binary catalyst — the August 25, 2026 PDUFA for first-line GEA — has already resolved positively. With the drug now approved for two indications and already commercializing, there is no hard dated binary catalyst in the next ~12 months. The next potential catalysts (colorectal cancer filing, Phase 3 data maturation) are not yet scheduled. Per the rubric, an approved drug with no pending regulatory decision scores 3-5; this is set at 4 given zanidatamab's meaningful but not sole contribution to Jazz's diversified $4.3B revenue base.
The PDUFA binary for GEA has just resolved positively (August 25, 2026). For JAZZ, watch commercial launch execution and the pace of Ziihera uptake in the GEA market. For ZYME, the $250M milestone is locked in, but the bigger story is the 10-20% royalty tail on Jazz's net sales — any positive sales updates or a colorectal cancer filing (which has Breakthrough Therapy Designation) could drive further upside. Both stocks have strong analyst support (JAZZ: 24 analysts, mean target $281, +13.5% upside; ZYME: 17 analysts, mean target $40.33, +32.7% upside).

Executive Summary
Zanidatamab (brand name Ziihera, development code ZW25) is a bispecific antibody targeting HER2-positive cancers, originally developed by Zymeworks (ZYME) and licensed to Jazz Pharmaceuticals (JAZZ). The drug received its second FDA approval on August 25, 2026 — this time for first-line HER2-positive unresectable locally advanced or metastatic gastroesophageal adenocarcinoma (GEA), adding to its prior accelerated approval in HER2-positive biliary tract cancer. The approval triggered a $250 million milestone payment from Jazz to Zymeworks, bringing total payments to $650 million, with up to $1.3 billion in additional milestones and 10-20% tiered royalties. The drug also holds Breakthrough Therapy Designation for HER2-positive colorectal cancer, suggesting further label expansion potential. For Jazz, zanidatamab is a meaningful growth driver within a diversified $4.3B-revenue portfolio; for Zymeworks, it is the central value driver with a royalty stream now beginning to materialize.
How It Works
Zanidatamab is a bispecific antibody — a type of engineered protein that grabs onto two different parts of the HER2 receptor on cancer cells at the same time. HER2 is a protein found on the surface of some cancer cells (like certain stomach, esophageal, bile duct, and breast cancers) that tells them to grow. By binding to two distinct spots on HER2, zanidatamab blocks growth signals more effectively than standard HER2-targeting drugs and also flags cancer cells for destruction by the immune system.
Trial Status
Zanidatamab has received two FDA approvals: (1) accelerated approval for HER2-positive biliary tract cancer, and (2) full approval on August 25, 2026 for first-line HER2-positive unresectable locally advanced or metastatic gastroesophageal adenocarcinoma (GEA), supported by Phase 3 HERIZON-GEA-01 trial data published in the New England Journal of Medicine. An ongoing Phase 3 trial (NCT05152147, n=920) is evaluating zanidatamab with chemotherapy ± tislelizumab in HER2-positive advanced gastric and esophageal cancers, with estimated primary completion in July 2026. A Phase 2 trial (NCT05027139) combining zanidatamab with evorpacept in HER2-expressing cancers has completed enrollment.
