- Why 4/10
- YORVIPATH is already FDA-approved and commercially launched with no pending regulatory decision or pivotal readout in the next ~12 months. The defining binary catalyst (FDA approval) has passed. Commercial ramp and sales growth are not binary catalysts. Per the rubric, an approved drug with no imminent regulatory decision scores 3-5, and YORVIPATH lands at 4 — it is the primary value driver for Ascendis but lacks a hard dated catalyst.
YORVIPATH is already approved and generating substantial revenue, so the binary approval catalyst has passed. Investors should focus on the commercial ramp trajectory (quarterly revenue growth, patient enrollment trends, and international expansion) and watch for any label-expansion catalysts (pediatric hypoparathyroidism). The stock trades at ~$263 with a consensus price target of ~$314 (20% upside) and a Strong Buy analyst consensus.

Executive Summary
YORVIPATH (palopegteriparatide, developed as TransCon PTH) is a once-daily parathyroid hormone replacement therapy approved by the FDA in August 2024 as the first and only treatment for hypoparathyroidism in adults. Developed by Ascendis Pharma (Nasdaq: ASND), the drug uses the company's TransCon prodrug technology to deliver continuous PTH exposure. YORVIPATH has seen a rapid commercial launch with Q2 2026 revenue of €252 million (up 145% YoY from €103M), is available in over 35 countries, and has enrolled more than 5,300 U.S. patients. With the only prior competitor (NATPARA) discontinued, YORVIPATH holds a market monopoly in a rare disease with no other approved alternatives.
How It Works
YORVIPATH is a hormone replacement therapy that replaces the missing parathyroid hormone (PTH) in patients with hypoparathyroidism. It is a prodrug — an inactive form that becomes active in the body — of teriparatide (a fragment of human PTH), delivered via a once-daily injection. Ascendis' TransCon technology attaches the drug to a carrier molecule that slowly releases active PTH over 24 hours, providing stable, continuous hormone levels to normalize calcium and phosphate metabolism.
Trial Status
YORVIPATH was approved by the FDA on August 12, 2024 (original NDA approval August 9, 2024) based on the global Phase 2 PaTH Forward and Phase 3 PaTHway trials. Long-term follow-up data from the Phase 3 PaTHway trial through Week 182 (3.5 years) showed 86% of patients met the multi-component efficacy endpoint, with sustained renal, bone, and quality-of-life benefits and no new safety signals. The drug was previously approved in the EU in November 2023.
The Two-Sided Case
