- Already approved and marketed (September 2023) for mydriasis reversal. Presbyopia label expansion decision expected October 17, 2026.
- Why 7/10
- The next hard catalyst is the FDA PDUFA date of October 17, 2026, for the supplemental NDA seeking to expand the label to include presbyopia — a large-market indication (an estimated 128 million Americans affected). This is a binary regulatory decision ~3.5 months out on an already-approved molecule that is the company's only current revenue source via the Viatris partnership, though the gene therapy pipeline (especially OPGx-LCA5) is the primary investment thesis.
Watch for the October 17, 2026 PDUFA decision on the presbyopia sNDA — approval would unlock a much larger addressable market via Viatris' commercial infrastructure. The stock's 95%+ implied upside to the $10.56 mean analyst target reflects optimism on both the presbyopia expansion and the gene therapy pipeline; a CRL or delay would pressure shares. Also monitor LYNX-2 (night vision) filing plans and OPGx-LCA5 Phase 3 dosing start in Q4 2026.

Executive Summary
RYZUMVI (phentolamine ophthalmic solution 0.75%) is an FDA-approved eye drop for reversing pharmacologically induced pupil dilation. It was developed by Ocuphire Pharma, which acquired Opus Genetics in October 2024, renamed the combined entity Opus Genetics, and now trades as IRD on Nasdaq. The drug is marketed in the US by Viatris. Beyond the approved indication, RYZUMVI has positive Phase 3 data in presbyopia (VEGA-3, June 2025) and night vision disturbances after LASIK (LYNX-2, June 2025). The FDA accepted a supplemental NDA for presbyopia with a PDUFA goal date of October 17, 2026. If approved, RYZUMVI would enter a growing but competitive presbyopia eye-drop market currently led by AbbVie's Vuity (pilocarpine) and newer entrants Qlosi and Yuvezzi. Opus Genetics' primary value driver, however, is its gene therapy pipeline for inherited retinal diseases; RYZUMVI provides revenue through the Viatris cost-sharing and royalty arrangement ($14.2M in 2025).
How It Works
An alpha-adrenergic blocker eye drop that shrinks the pupil back to normal size. For the already-approved use, it reverses dilation caused by exam drops. For presbyopia (age-related difficulty seeing up close), constricting the pupil increases depth of focus, improving near vision — similar to how a pinhole camera works.
Trial Status
FDA-approved (September 2023) for reversal of pharmacologically-induced mydriasis; commercially launched by partner Viatris in April 2024. sNDA filed and accepted for presbyopia (Phase 3 VEGA-2 and VEGA-3 trials both met primary endpoints) with PDUFA October 17, 2026. Phase 3 LYNX-2 trial for night vision disturbances after keratorefractive surgery also met its primary endpoint in June 2025.
