- Trial Phase
- BLA resubmission pending (accelerated approval) — prior BLA reached PDUFA Feb 8, 2026 and received a Complete Response L
- Time to Market
- Potential US approval in roughly Q1–Q2 2027 if the BLA is resubmitted in Q3 2026 and granted accelerated approval (a post-approval confirmatory trial would still be required); approval is not assured.
- Why 6/10
- Next hard dated catalyst: BLA resubmission for accelerated approval is on track for Q3 2026 (by ~Sept 30, 2026), and a Class-2 resubmission typically gets a ~6-month FDA review, so the real binary — the approval decision — should land roughly Q1–Q2 2027, inside the 12-month window; it is a major but not sole value driver (REGENXBIO also has RGX-202 and sura-vec readouts), hence 6.
Watch for the NAVSUNLI BLA resubmission before end of Q3 2026 and the FDA's acceptance/PDUFA date that follows (~6 months later) — that PDUFA, not the resubmission itself, is the binary that will swing RGNX. Note the stock has already re-rated from ~$5.70 (May 2026) to ~$12 (Aug 2026) on the FDA alignment, so much of the good news may be priced in; the July 2026 ~$108M offering (~17-20% dilution) and Barclays' Aug 7 downgrade to Equal-Weight are offsetting negatives.

Executive Summary
NAVSUNLI (clemidsogene lanparvovec, code RGX-121) is REGENXBIO's one-time AAV9 gene therapy for Hunter syndrome (MPS II) and would be the first gene therapy / first one-time treatment for the disease if approved. The FDA rejected the original BLA around its Feb 8, 2026 PDUFA date, then reversed course on June 22, 2026 and aligned on a path to resubmit for accelerated approval with no additional studies required; REGENXBIO guided the resubmission to Q3 2026 with a positive Type A meeting, implying a decision roughly 6 months later. The drug carries Orphan, Rare Pediatric Disease, Fast Track and RMAT designations and is partnered with Nippon Shinyaku, but it competes with Takeda's weekly-infusion enzyme therapy Avlayah, and REGENXBIO shares are volatile and diluted (a July 2026 ~$108M offering) with other big catalysts (RGX-202, sura-vec) competing for attention.
How It Works
NAVSUNLI is a one-time gene therapy for Hunter syndrome (MPS II), a rare X-linked disease in boys where a missing enzyme (I2S) causes progressive brain and body damage. A harmless AAV9 virus is delivered to the central nervous system carrying a working copy of the IDS gene, so the patient's own cells produce the enzyme permanently — versus today's standard of care, which is weekly intravenous enzyme infusions for life.
Trial Status
Pivotal clinical data were submitted in the original BLA. FDA issued a CRL around February 2026 (PDUFA was Feb 8, 2026) citing uncertainty over trial design; on June 22, 2026 FDA aligned on resubmitting for accelerated approval with no new control arm, no new enrollment, and no additional studies; REGENXBIO says the resubmission is on track for Q3 2026 after a positive FDA Type A meeting, with confirmatory evidence of clinical benefit required post-approval under the accelerated pathway.
The Two-Sided Case
Upcoming Catalysts
- NAVSUNLI BLA resubmission (on track for Q3 2026, by ~Sept 30, 2026)
- FDA acceptance of the resubmission and the resulting PDUFA target date (typically ~6 months after filing, so roughly Q1–Q2 2027)
- FDA approval decision on the resubmitted BLA
- Company-level (not NAVSUNLI-specific): RGX-202 Duchenne BLA submission in Q3 2026 and sura-vec wet AMD pivotal topline in Q4 2026
