- Trial Phase
- Phase 2
- Time to Market
- If Phase 2 succeeds and Phase 3 initiates in 2027, potential approval would likely be 2029-2030 at the earliest, assuming positive pivotal data.
- Why 7/10
- The next hard, dated catalyst is the NAVIGATE-2 Phase 2 topline data readout expected in H1 2027 (~3-8 months away). MIL116/CLYM116 is Climb Bio's lead asset and the company is essentially a single-product IgAN story, so this binary readout will drive the vast majority of share-price movement. ImpactScore 7 reflects a pivotal readout within ~6-12 months on the company's lead asset.
The next major catalyst is NAVIGATE-2 Phase 2 topline data in H1 2027. If data replicates Phase 1's best-in-class profile (Q12W dosing, ~33-day half-life, >90% APRIL suppression), the stock could re-rate significantly toward the $27.60 mean analyst target. Conversely, any safety or efficacy miss in actual IgAN patients would be severely punished given the single-asset concentration.

Executive Summary
MIL116 (also known as CLYM116) is Climb Bio's lead asset — an anti-APRIL monoclonal antibody in development for IgA nephropathy (IgAN), a chronic kidney disease with no cure. Phase 1 data presented at ASN Kidney Week 2026 showed a best-in-class ~33-day half-life, >90% APRIL suppression, and ~70% reduction in the key disease biomarker Gd-IgA1 with a single 800 mg dose, supporting a convenient every-12-week dosing regimen. The drug was well tolerated with no serious adverse events across 88 participants. The Phase 2 NAVIGATE-2 trial is enrolling IgAN patients with initial data expected in H1 2027, and a global Phase 3 trial is planned for 2027. Climb Bio trades on Nasdaq under CLYM at ~$10.19 with a ~$624M market cap, and analysts are overwhelmingly bullish (20 Buy/Strong Buy vs. 1 Hold) with a mean price target of $27.60, implying ~175% upside.
How It Works
MIL116 is a monoclonal antibody that blocks APRIL, a signaling protein that stimulates the production of abnormal IgA antibodies. In IgA nephropathy (IgAN), these abnormal antibodies build up in the kidneys and cause inflammation and damage. By neutralizing APRIL, MIL116 reduces production of the disease-causing antibodies, potentially slowing or stopping kidney damage. Its long half-life (~33 days) allows for injections once every 12 weeks, which is more convenient than many competing drugs.
Trial Status
Phase 1 completed in healthy volunteers (n=88) with positive data reported at ASN Kidney Week 2026. Phase 2 NAVIGATE-2 trial is actively enrolling patients with IgAN. Global Phase 3 registrational trial planned for 2027 pending regulatory feedback.
The Two-Sided Case
