- Trial Phase
- Phase 3 (completed)
- Time to Market
- Targeting potential US/EU commercial launch in 2027, pending regulatory filings and review
- Why 5/10
- Bota-vec has completed Phase 3 and MeiraGTx plans to file BLA/MAA immediately (manufacturing validation complete), targeting a potential 2027 launch. However, there is no specific PDUFA date set yet — the next catalyst is the regulatory submission itself, which is imminent but undated. This caps the impactScore at 5 per the rubric (no dated binary within ~6 months), despite bota-vec being a major value driver for MGTX.
Watch for MeiraGTx's BLA submission to the FDA for bota-vec — that filing, and its acceptance, will be the next major catalyst. The $400M Oberland Capital financing (announced July 7, 2026) removes near-term dilution risk and funds launch preparations. The key risk: regulators may view the Phase 3 LUMEOS primary endpoint miss as problematic, even though secondary endpoints were positive.

Executive Summary
Botaretigene sparoparvovec (bota-vec) is a late-stage AAV5-based gene therapy for X-linked retinitis pigmentosa (XLRP), a rare inherited retinal disease causing blindness with no approved treatments. MeiraGTx reacquired full rights from Johnson & Johnson in April 2026 for $25M upfront after the Phase 3 LUMEOS trial missed its primary endpoint but showed positive secondary visual-function results. The company plans immediate global regulatory filings (US, EU, Japan) with manufacturing already validated and a potential 2027 commercial launch. Bota-vec is a key value driver for MGTX alongside other pipeline programs, supported by recent $400M Oberland Capital financing. The mixed Phase 3 data creates regulatory uncertainty but the drug benefits from FDA Fast Track and Orphan Drug designations.
How It Works
A one-time gene therapy that uses a harmless virus (AAV5) to deliver a working copy of the RPGR gene directly into cells of the retina (the light-sensitive tissue at the back of the eye). In XLRP, mutations in the RPGR gene cause progressive vision loss and eventual blindness, primarily in young males. By restoring the missing protein, the therapy aims to halt or reverse retinal degeneration.
Trial Status
The Phase 3 LUMEOS trial (NCT04671433) completed with mixed results: the novel primary endpoint (vision-guided mobility through a maze) was not met. However, multiple secondary endpoints were positive, including a statistically significant improvement in low luminance visual acuity (p=0.003), with 45% of treated patients gaining 10+ letters vs. control. The Phase 1/2 MGT009 study previously demonstrated safety and vision improvement. Manufacturing process performance qualification (PPQ) is complete, meaning commercial supply is ready.
The Two-Sided Case
