- Trial Phase
- Phase 3
- Time to Market
- ~3-4 years if Phase 3 succeeds (topline H2 2027, then FDA filing/approval ~2028-2029)
- Why 5/10
- The next hard dated binary is the Phase 3 CoMpass topline data (15-month primary endpoint) expected in H2 2027 — more than 12 months away, capping the score at 5. Bel-sar drives essentially all of Aura's valuation, so it sits at the high end of that cap. Earlier near-term data from the Phase 1/2a bladder cancer program (expected mid-2026) is non-pivotal and unlikely to be a company-defining binary.
Wait for the H2 2027 Phase 3 topline readout as the primary catalyst. The stock may drift in the interim on bladder cancer data points and pipeline updates. Recent heavy dilution ($299M offering at $6.00) resets the base, but the company is now well-funded through H2 2028. Analyst consensus is strongly bullish (11:1 buy-to-hold) with a mean $18.80 target (~144% upside from ~$7), though that reflects the binary optionality of Phase 3 success.

Executive Summary
Bel-sar (belzupacap sarotalocan / AU-011) is Aura Biosciences' lead and only late-stage asset — a first-in-class virus-like drug conjugate (VDC) being developed as a vision-sparing first-line therapy for early choroidal melanoma, a rare eye cancer. Phase 2 results showed 80% tumor control and 90% vision preservation, which supported the design of the Phase 3 CoMpass trial under a Special Protocol Assessment (SPA) with the FDA. Enrollment of 108 patients completed in June 2026, with topline data expected in H2 2027 — the stock's defining binary event. The company also has early-stage data in bladder cancer (NMIBC) showing promising complete response rates. A $299M equity offering in May 2026 funded the company through H2 2028 but massively diluted existing shareholders. The stock trades at ~$7 with a consensus analyst target of $18.80, reflecting strong confidence in the Phase 3 data but with significant execution risk still ahead.
How It Works
Bel-sar is a new kind of cancer drug called a "virus-like drug conjugate" (VDC). Think of it as a stealth delivery system: a hollow shell made from a harmless virus protein (like a decoy) carries a light-activated toxin. It's injected near the tumor. The virus shell seeks out and binds to cancer cells. Then the doctor shines a specific wavelength of near-infrared laser light on the area, which activates the toxin to rapidly destroy the cancer cells. It also appears to stimulate the immune system to fight remaining tumor cells. The approach aims to kill tumors while preserving healthy surrounding tissue — especially important in the eye.
Trial Status
Phase 3 CoMpass trial fully enrolled (108 patients, June 2026). Topline data from the 15-month primary endpoint expected in the second half of 2027. Phase 2 end-of-study data showed 80% tumor control and 90% vision preservation. Phase 1 bladder cancer trial completed with 4/5 low-grade patients achieving complete response. Phase 1/2a NMIBC trial ongoing.
The Two-Sided Case
Upcoming Catalysts
- Mid-2026: 3-month NMIBC (bladder cancer) data update from ongoing Phase 1/2a trial
- H2 2027: Phase 3 CoMpass topline 15-month primary endpoint data in choroidal melanoma — the defining binary event for the stock
- Potential FDA/EMA regulatory milestones and interactions given Fast Track and Orphan Drug designations
